Effect of a mouthwash based on propolis against Streptococcus mutans and Porphyromonas gingivalis

Abstract

Objective: To characterize chemically an ethanolic extract derived from propolis from Oxapampa city, Peru; Additionally, develop an experimental mouthwash based on the ethanolic extract of propolis and demonstrate its antibacterial activity against Streptococcus mutans ATCC 25175 and Porphyromonas gingivalis ATCC 33277.

Materials and methods: Experimental type research, in vitro. A phytochemical march was done, followed by high-performance liquid chromatography (HPLC) for chemical analysis. Antibacterial sensitivity was evaluated by Kirby Bauer's method and Kruskall-Wallis test followed by Dunn's post hoc and Mann-Witney U test were used to compare the effect of propolis extract on the strains. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined. To compare the inhibition effect of the mouthwash between each strain, Student's t-test was applied. The significance level adopted was p < 0.05.

Results: Propolis from Oxapampa city, Peru, evidenced the presence of gallic acid (10.19 ?g/mL) and chlorogenic acid (412.89 ?g/mL). The minimum bactericidal concentration of the ethanolic extract of propolis was 30.537 mg/mL The mouthwash based on the extract formed larger inhibition halos (p<0.05) against S. mutans ATCC 25175 (29.7 mm) and P. gingivalis ATCC 33277 (28.39 mm) than 0.12% chlorhexidine (18.28 mm).

Conclusions: There are differences between the antibacterial effect of the mouthwash based on ethanolic extract of propolis and 0.12% chlorhexidine against Streptococcus mutans ATCC 25175 and Porphyromonas gingivalis ATCC 33277.
